Is there a discrepancy between the chemical and and microscopic methods for detection of WBC's and if so why is this happening? Answer: Yes, there is a discrepancy between the two methods. The leukocyte esterase stix method is reading negative, while the microscopic results show 50-100 WBC/hpf. This pattern is often seen in kidney transplant rejection. The wbc's that are responsible for the rejection are the B and T lymphocytes. However the chemical Leukocyte esterase method only can detect neutrophils, eosiniphils and basophils. The B and T lymphocytes can still be seen under the microscope.
